Estas intera\u00e7\u00f5es recrutam a PI3K para seu substrato, o fosfatidil-inositol 4,5-bifosfato (PIP2) permitindo a gera\u00e7\u00e3o do segundo mensageiro lip\u00eddico&nbsp;fosfatidil-inositol 3,4,5-trifosfato (PIP3). A Akt e a PDK1 s\u00e3o recrutadas para a membrana plasm\u00e1tica pela associa\u00e7\u00e3o com PIP3. Isto permite que a Akt seja ativada atrav\u00e9s da fosforila\u00e7\u00e3o no res\u00edduo Thr-308 pela PDK1 e Ser-473 pelo mTORC2. Uma vez ativa, a Akt fosforila muitos alvos, inclu\u00edndo m\u00faltiplos s\u00edtios na TSC2, que forma um complexo funcional com TSC1. A fosforila\u00e7\u00e3o de TSC2 prejudica a habilidade de o complexo TSC1-TSC2 atuar na GTPase Rheb, resultando no ac\u00famulo de Rheb-GTP.<\/div>\n<div class=\"title\" style=\"color:#212121;font-family:Arial, sans-serif;font-size:15px;line-height:22px;text-align:justify\">Atrav\u00e9s de um mecanismo ainda pouco conhecido, a Rheb-GTP ativa a mTORC1, que fosforila e inibe a 4E-BP1 e ativa a S6K1 e S6K2. Um <i>loop<\/i>&nbsp;de retroalimenta\u00e7\u00e3o negativa existe, no qual o mTORC1 e S6K1 fosforilam diretamente o IRS-1 e bloqueiam a sinaliza\u00e7\u00e3o da insulina e IGF-1 atrav\u00e9s da PI3K. Fatores de crescimento tamb\u00e9m aumentam a atividade da mTORC2, apesar de os eventos moleculares para tal ainda n\u00e3o serem bem compreendidos. Atrav\u00e9s de um mecanismo distinto, o complexo TSC1-TSC2 pode se ligar ao complexo mTORC2, ativando-o. Al\u00e9m disso, o complexo TSC1-TSC2 parece estar envolvido na fosforila\u00e7\u00e3o da PKC-alfa pela mTORC2. PDK1 fosforila S6K1, S6K2 e PKC-alfa de uma maneira independente de PIP3.
